In order to explore the endophytic bacteria resources of soybean roots in soda saline-alkali land, the soybean roots in Daqing soda saline-alkali land were used as materials in this study. The endophytic bacteria in soybean roots were isolated, purified and identified by tissue grinding method and plate streaking method, and their phosphate solubilization, nitrogen fixation, siderophore production, ACC deaminase, IAA ability and soda saline-alkali tolerance were determined. The germination experiment was carried out under soaking conditions. The results showed that the three strains had the ability of phosphate solubilization, nitrogen fixation, siderophore production, plant hormone IAA production and ACC deaminase production. Moreover, the three strains also had strong resistance to soda saline-alkali, which could tolerate 100 ~ 150 mmoL . L-1 soda salt ( NaHCO3 and Na2CO3, molar ratio of 9 : 1 ), and the maximum pH tolerance could reach 8.8. The germination experiment of three root endophytic bacteria found that the three root endophytic bacteria could significantly increase the number of root tips, root length, root surface area and volume after soaking the seeds of two varieties of soybean seeds. It is proved that the three endophytic bacteria from soybean root have the function of promoting growth and have the potential as biological bacterial fertilizer. |
