为了使EMS诱变在谷子种质资源创新中发挥最佳效果,获得优异的变异资源,本研究对谷子新品种‘公谷88’进行了3种浸种时间、3种EMS浓度的化学诱导,并对M1种子发芽及幼苗生长的诱变效应进行研究。结果表明:与对照相比,低剂量EMS处理(0.4 %,6 h)对公谷88的发芽势、发芽率以及幼苗生长影响差异不显著,随着EMS浓度升高及处理时间延长,公谷88萌发及幼苗生长受到逐渐抑制,1.2%EMS浓度浸种14h对公谷出苗及幼苗生长影响最严重,大大降低发芽势、发芽率、成苗率,幼苗生长受到严重抑制。以成苗率达到半致死剂量为标准,通过回归方程计算出在浸种时间6 h、10 h、14 h下EMS处理浓度的半致死剂量分别为1.01 %、0.75 %、0.39 %,可作为EMS诱变公谷88建立突变体库的适宜条件。 |

In order to make EMS mutation play the best effect in the innovation of foxtail millet germplasm resources and obtain excellent mutation resources, this study conducted three kinds of seed soaking time and three kinds of chemical induction of EMS concentration on a new millet variety‘Gonggu 88’, the mutagenic effects of M1 seed Germination and seedling growth were studied.The results showed that,compared with control, low dose EMS treatment (0.4% , 6 h) had no significant effect on germination potential, germination rate and seedling growth of Gonggu 88. With the increase of EMS concentration and treatment time, germination and seedling growth of Gonggu 88 were gradually inhibited,Seed soaking with 1.2% ems concentration for 14 h had the most serious effect on the seedling emergence and growth of Gonggu. The germination potential, germination rate and seedling rate were greatly reduced, and the seedling growth was severely inhibited.According to the standard of semi-lethal dose, the semi-lethal dose of EMS treatment was 1.01% , 0.75% and 0.39% at 6 H, 10 H and 14 h soaking time, respectively, it can be used as the suitable condition for EMS mutagenesis to establish the mutant library of Gonggu 88. |
