采用水培试验,以PEG 6000为渗透剂,研究了渗透胁迫对烤烟内源多胺含量及其代谢酶活性的影响。结果表明,渗透胁迫处理使烤烟根中腐胺(Pu t)、精胺(Spm)和亚精胺(Spd)含量显著增加,而且随渗透势降低变化幅度增大;随胁迫时间延长,Pu t的含量先升高后下降,而Spm和Spd含量在胁迫处理6 h时最高,之后逐渐降低。渗透胁迫下烤烟叶片中Pu t含量和精氨酸脱羧酶(ADC)活性的变化趋势与根中Pu t含量的变化相一致;Spd含量和多胺氧化酶(PAO)活性均在胁迫处理18 h达到高峰,而后显著下降;Spm含量和鸟氨酸脱羧酶(ODC)活性没有明显变化。叶片中Pu t含量与其ADC活性呈显著正相关,提示渗透胁迫下烤烟叶片Pu t的合成主要是由精氨酸在ADC的作用下直接脱羧而形成的,而Pu t的积累有利于提高烟株的抗旱性。 |

The effect of osmotic stress on endogenous polyamines content and their metabolism enzyme activity in tobacco plants were investigated by hydroponic experiment using polyethylene glycol(PEG6000) as osmoticum.The results showed that the contents of putrescine(Put),spermidine(Spd) and spermine(Spm) in tobacco roots increased significantly under osmotic stress,and with the decreasing of osmotic potential,the changing ranges of these polyamines were more significant compared with control.It was also found that the content of Put in the roots increased during osmotic stress for 12 h at the osmotic potential of-0.5 MPa and 18 h at the osmotic potential of-0.25 MPa,respectively;afterwards,the Put content in two stress-treated roots all decreased.The contents of Spm and Spd in the roots were the highest with osmotic stress for 6 h,after that they gradually declined.The changing trends of Put content and ADC activity in the leaves were compatible with Put content in the roots,while both the Spd content and PAO activity reached their peaks with osmotic stress for 18 h,then decreased obviously.There were not significant changes in Spm content and ODC activity in the leaves during osmotic stress.In addition,it was found that there was a significant positive relationship between Put content and ADC activity in the leaves,which suggested that the major pathway of Put biosynthesis in the leaves under osmotic stress was from arginine decarboxylation by the function of ADC,and the accumulation of Put in the plants could enhance the drought-resistance of tobacco. |
