通过对鹰嘴豆籽粒中的异黄酮提取和测定方法的优化,拟建立一套适用于测定鹰嘴豆总异黄酮的三波长紫外分光光度法。结果表明:鹰嘴豆总异黄酮最佳提取条件为:0.30 g不脱脂豆粉溶于25 ml浓度70%的乙醇溶液中,在60℃水浴温度下提取3 h;一次提取效率为86.5%,两次合并提取效率可达97.0%;建立的测定方法精密度变异系数(C V)为1.14%,平均加标样回收率为99.0%,变异系数(C V)为3.2%。采用三波长测定可有效消除干扰物质对测定的影响,该方法具有操作简便、重现性好、精密度高等特点,适于大批量样品测定以及产品生产的质量控制及监测。 |

The extraction and determination were optimized to build a determining method of the total isoflavone content of chickpea seeds by Three-wavelength UV spectrophotometry.The optimum extraction condition was: 0.3 g chickpea flour was extracted with 25 ml 70% alcohol density for 3 hours.The extracting temperature was 60℃.The isoflavone extracting rate was 86.5% at one time,while the total extraction rate of two times was 97.0%.The coefficient of variation(CV) of the accuracy experiment was 1.14%.The average recovery rate was 99.0% with CV=3.2%.The newly developed determining method is reliable,simple and accurate,and could avoid or eliminate the influence of the interfering components and the peak floating,and is suitable for the quality control of chickpea products. |
