0℃冻害分别处理蚕豆幼苗0、3、6、12 h,测定了蚕豆幼苗叶片的蛋白质、脯氨酸、还原性谷胱甘肽和丙二醛等生理指标,进行了蚕豆低温胁迫下的RAP-PCR指纹分析和抵抗低温冻害的特异基因序列片段的克隆。结果表明:随着低温胁迫的加强,蚕豆幼苗叶片内蛋白质含量呈现先升后降的趋势,脯氨酸相对百分含量逐渐增加,还原性谷胱甘肽含量逐渐下降,丙二醛含量低温胁迫后维持在一个较低的稳定水平。以8条随机引物进行RAP-PCR指纹分析鉴定出8条抵抗冻害的特异性片段,并成功克隆到其中的4条。 |

Protein content, proline, reduced glutathione, malondialdehyde of Vicia faba at seedling stage were measured in 0℃, RAP-PCR fingerprint analysis was conducted and chilling damage-resistance gene segments were cloned. The results showed that proline increased with the time at 0℃, reduced glutathione decreased with th
e time, protein content increased first but decreased then, and malondialdehyde stayed at a steady level after freezing injury. Eight chilling damage-resistance genes were identified with 8 random primers and RAP-PCR, and four of them were successfully cloned. |
