      对卫星搭载苜蓿种子的叶片进行愈伤组织的诱导和NaCl、PEG及温度胁迫处理,探讨空间环境对不同逆境下苜蓿愈伤组织的脯氨酸、可溶性糖、可溶性蛋白、相对电导率和丙二醛含量5项生理指标的影响,采用隶属函数法对搭载愈伤组织的抗逆性进行评价。结果表明:未搭载材料经NaCl(2%)、PEG(30%)、高温(40℃,6 d)和低温(0℃,8 d)胁迫后,其存活率分别为2.68%,3.8%,43.68%和35.42%,与未胁迫组间存在极显著差异。4份苜蓿愈伤组织中,搭载的苜蓿愈伤组织在脯氨酸、可溶性糖、可溶性蛋白含量均高于未搭载愈伤组织,相对电导率和丙二醛含量均低于未搭载愈伤组织。搭载的苜蓿愈伤组织比未搭载的愈伤组织的丙二醛含量降低了39.47%,相对电导率降低了48.37%,可溶性糖含量增加了34.71%,可溶性蛋白含量增加了40.67%,脯氨酸的含量增加了55.6%。搭载的苜蓿愈伤组织抗逆性强弱为:干旱>盐>低温>高温,卫星搭载胁迫后的苜蓿愈伤组织对干旱和盐的抵抗能力比较强。
英文摘要:
      To study the influence of space environment on the leaf callus of alfalfa seed carried by satellite, the callus of alfalfa under different stress (NaCl and PEG and temperature stress) were studied, and the five physiological parameters including proline, soluble sugar, soluble protein, relative conductivity and MDA content were determined to evaluate the effect of satellite carrying on callus resistance. The results showed that the survival rates of no-carrying material were 2.68%, 3.8%, 43.68%, and 35.42% for NaCl (2%), PEG (30%), high temperature (40℃, 6 d), and low temperature (0℃, 8 d) stress, being significantly different with those of the control. Satellite-carrying resulted in higher proline, soluble sugar, protein contents, by an increase of 55.6%、34.71%, and 40.67%, respectively. By contrast, the relative conductivity and MDA content were reduced by 48.37% and 39.47%. The degree of resistance of satellite carrying alfalfa callus was in the order of drought, salt, low temperature, and high temperature.
