为了研究紫花苜蓿(Medicago Sativa L.)对辐射和高温胁迫的生理响应,分别以800 Gy和600 Gy 60Co-γ辐射苜蓿种子的第二代高株变异植株和未辐射苜蓿种子植株叶片愈伤组织的分化苗(G3,G4,CK)为材料,模拟40℃高温胁迫(时间为1,2,3,4 d),测定高温胁迫下苜蓿分化苗存活率及8项抗性生理指标,对8项指标进行隶属函数和综合D值计算,综合分析3组分化苗的耐热能力。结果表明:CK分化苗在40℃高温胁迫4 d存活率为64%,胁迫温度高于40℃和超过4 d严重影响分化苗存活率,确定有效模拟高温胁迫为40℃ 4 d;40℃高温胁迫下,3组分化苗可溶性蛋白、可溶性糖、脯氨酸含量和过氧化物酶、超氧化物歧化酶活性均随胁迫时间的增加呈先升后降趋势,过氧化氢酶活性呈降低趋势,丙二醛含量和相对电导率呈增加趋势,其中G3分化苗在高温胁迫3 d,可溶性蛋白、可溶性糖、脯氨酸、丙二醛的含量、相对电导率和过氧化物酶、超氧化物歧化酶活性比其对照分别增加了235.74%,79.21%,323.65%,30.10%,18.09%,82.87%和350.91%,过氧化氢酶活性比其对照降低了64.52%。8项生理生化指标的综合D值分析表明,3组分化苗的耐热能力排序为G3>G4>CK,一定剂量辐射处理可提高苜蓿分化苗的抗性。 |

To study the physiological response of alfalfa (Medicago sativa L.) under radiation and heat stress, the differentiated seedlings of alfalfa callus were used as materials. The treatments G3 and G4 of second generation tall plants were differentiation of leaf callus of irradiated alfalfa seed of 800 Gy and 600 Gy variant plants and CK was non irradiated alfalfa seed plants. The differentiated seedlings were subjected to high temperature stress at 40 ℃ for 1, 2, 3 and 4 days to investigate the survival rate and physiological and biochemical characteristics of alfalfa differentiation seedlings. The membership function and comprehensive D value of 8 indexes were calculated to comprehensively analyze the heat resistance of three groups of differentiated seedlings. The experimental results indicated that the survival rate of control differentiated seedlings was 64% under heat stress at 40℃ for 4 days. The survival rate of differentiated seedlings was seriously affected when the stress temperature was higher than 40℃ for more than 4 days. The effective simulated heat stress was 40℃ for 4 days. Under 40℃ heat stress, the contents of soluble protein, soluble sugar, proline and the activities of peroxidase and superoxide dismutase in three groups of differentiated seedlings increased first and then decreased with the increase of stress time, the activity of catalase decreased, and the content of malondialdehyde and relative conductivity increased. Under high temperature stress for 3 days,the contents of soluble protein, soluble sugar, proline, malondialdehyde, relative conductivity, peroxidase, and superoxide dismutase activities of G3 differentiated seedlings increased by 235.74%,79.21%,323.65%,30.10%,18.09%,82.87% and 350.91% respectively, and the catalase activity decreased by 64.52%. The comprehensive D value analysis of 8 physiological and biochemical indexes showed that the heat resistance of the three groups of differentiated seedlings was G3 > G4 > CK. A certain dose of radiation treatment improved the stress resistance of alfalfa differentiated seedlings. |
