酸枣(Ziziphus jujuba var. spinosa )具有优良的抗旱性,MYB转录因子在植物应对干旱胁迫时发挥重要作用。为探究酸枣中MYB转录因子的抗旱机理,本研究通过生物信息学方法对酸枣MYB基因家族进行鉴定分析,结合干旱胁迫下的转录组数据,以期筛选响应干旱胁迫的关键基因,为抗旱优质酸枣品种的培育提供关键候选基因。结果表明,在酸枣全基因组鉴定获得62个MYB基因家族成员,分布在12条染色体上,均包含保守Motif2结构域,主要位于细胞质、细胞核及叶绿体中;酸枣、冬枣、骏枣的MYB成员可分为7个亚组;在酸枣全基因组内共鉴定出4361对共线性基因对,其中MYB家族成员中有4对共线性基因对;酸枣与骏枣和冬枣的MYB基因家族分别存在6对和10对共线性基因;在酸枣MYB基因家族的启动子区域鉴定出光响应、环境胁迫、激素响应等相关的41类顺式作用元件, 包括响应干旱胁迫的脱落酸响应元件ABRE和MYB结合位点元件MBS;在干旱胁迫的表达模式下,MYB成员ZjMYB14、ZjMYB21、ZjMYB26、ZjMYB43、ZjMYB51、ZjMYB53在枣不同发育时期均显著表达,可能为关键抗旱基因。 |

Ziziphus jujuba var. spinosa has excellent drought resistance, and MYB transcription factors play an important role in plant response to drought stress. In order to investigate the drought-resistant mechanism of MYB transcription factors in Ziziphus jujuba var. spinosa, the present study analyzed the identification of the MYB gene family of Ziziphus jujuba var. spinosa by bioinformatics, and combined with the transcriptome data under drought stress, in order to screen key genes responding to drought stress, and to provide key candidate genes for the cultivation of high-quality Ziziphus jujuba var. spinosa varieties that are drought-resistant. The results showed that 62 MYB gene family members were identified in the whole genome of Ziziphus jujuba var. spinosa, distributed on 12 chromosomes, and all of them contained the conserved Motif2 structural domain, which was mainly located in the cytoplasm, nucleus and chloroplasts; the MYB members of Ziziphus jujuba var. spinosa, Ziziphus jujuba Mill.cv.Dongzao, and Ziziphus jujuba Mill.cv.Junzao were classified into seven subgroups; In the whole genome of Ziziphus jujuba var. spinosa, 4361 gene pairs were identified, of which 4 pairs of MYB family members were identified; 6 pairs and 10 pairs of gene pairs existed in the MYB gene family of Ziziphus jujuba var. spinosa and Ziziphus jujuba Mill.cv.Junzao, and Ziziphus jujuba Mill.cv.Dongzao, respectively; in the promoter region of the MYB gene family of jujube, 41 types of cis-acting elements related to light response, environmental stress, hormone response and so on, were identified, including abscisic acid-responsive element ABRE in response to drought stress and MYB-binding site element MBS; in the drought-stress expression mode In the expression pattern of drought stress, the MYB members ZjMYB14, ZjMYB21, ZjMYB26, ZjMYB43, ZjMYB51, ZjMYB53 were significantly expressed in different developmental periods of jujube, which might be the key drought-resistant genes. |
