| 为探究蒙古黄芪根瘤内非共生细菌的分布类型及遗传多样性,采集内蒙古东部地区土壤种植蒙古黄芪,无菌条件下分离纯化根瘤内生细菌,基于16S rDNA基因序列构建系统发育树,分析基因多态性和菌株生物学特性。结果发现,蒙古黄芪根瘤内分离纯化得到12株菌,分别归属于根瘤菌属(Rhizobium,4株)、假单胞菌属(Pseudomonas,6株)和中慢生根瘤菌属(Mesorhizobium,2株),其中内蒙古根瘤菌(R. mongolense)和无花果假单胞菌(P. ficuserectae)为优势菌种。假单胞菌属与根瘤菌属、中慢生根瘤菌属间遗传距离(Dxy)分别为0.19110和0.17593,基因交流指数(Nm)远远小于1.0,彼此间未发生基因交流现象;假单胞菌属在3.0%NaCl、弱酸(pH=5)、强碱(pH=9)环境中OD600均高于1.0,表现出较强的耐盐、耐酸、耐碱性,可为内蒙古东部地区瘠薄沙土种植蒙古黄芪提供优良菌株,为共生菌和促生菌联合菌剂开发提供良好生物材料。 |

| To explore the distribution type and genetic diversity of non\|symbiotic bacteria in the nodules of Astragalus mongholicus Bunge, the soil in eastern Inner Mongolia was collected to plant A. mongholicus, and the endophytic bacteria in root nodules were isolated and purified under aseptic conditions. The phylogenetic tree was constructed based on the 16S rDNA gene sequence, and the gene polymorphisms and biological characteristics of the isolates were analyzed. The results showed that 12 isolates were isolated and purified from the nodules of A. mongholicus, which belonged to Rhizobium (4 strains), Pseudomonas (6 strains), and Mesorhizobium (2 strains). Among them, R. mongolense and P. ficuserectae were the dominant species. The pairwise genetic distance (Dxy) between Pseudomonas and Rhizobium, and Mesorhizobium were 0.19110 and 0.17593, respectivley. The gene exchange index (Nm) was far less than 1.0, and no gene exchange occurred between them. The OD600 values of Pseudomonas in 3.0% NaCl, weak subacidity (pH=5), and strong alkali (pH=9) environments were all higher than 1.0, exhibiting strong salt, acid, and alkaline tolerance. Therefore, they were the excellent strains exploited for cultivation of Astragalus mongholicus Bunge inoculated by combined microbial agents of symbiotic and growth\|promoting bacteria in eastern Inner Mongolia with poor sandy soil. |
