cDNA-AFLP(cDNA Amplified Fragment Length Polymorphism) was used to analyze genes differential expressed in broomcorn millet(Panicum miliaceum L.) seedling under the condition of normal watering,serious drought and rehydration.All of the thirty-six primer pairs used in the selective amplification could amplify differential expression profile among the samples.Twelve fragments only presented in serious drought or rehydration were selected,cloned and sequenced.Blastn search to NCBI dbEST showed that only two of the fragments had significant homologous nucleotide sequence with wheat and rice ESTs,others could not found significant homologous nucleotide sequence.Based on the Blastx search to NCBI non-redundant protein database,one fragment was similar to UDP-N-acetylglucosamine-N-acetylmuramyl-(Pentapeptide),two fragments had significant homologous protein sequence with rice retrotransposon,another two had significant homologous protein sequence with two hypothetical proteins. |
