Abstract:
      We used the different wheat seedings which were planted on the condition that water is restricted and resupply.The concentrated betain samples were purified by passing through the ion-exchange resin.We used the Reverse-Phase High Performance Liquid Chromatography technology to determine the change of betaine content.In the HPLC system, the buffer is 0.05 M sodium phosphate (pH4.45) , the column was kept at 28℃ and the flow rate was 0.6 mL/min, the samples was determined at 192 nm wavelength.Besides, we optimized the factors, such as temperature, time and ion pair.During both the PEG-induced drought stress and water resupplied process, the amount of betaine increased at first and reduced latter.A great deal of Betaine accumulated in the anti-drought type wheat in a short time.This result showed that the synthesis of betaine have close connection with anti-drought plant.This method was exact, sensitive and convenient, as well provided a basis for anti-drought mechanism and the wheat breeding work.
