Abstract: |
Callus of Glycyrrhiza uralensis was used in this research for extraction of total proteins, SDS-PAGE, loading quantity and staining method, allowing optimization of the two-dimensional gel electrophoresis (2D-E) technology. The results showed that improved acetone precipitation could increase the protein contents and image resolution of gel electrophoresis. There were 1 132 protein spots identified using silver staining when the loading quantity of the sample protein was 450 μg/(17 cm IPG trip). This research developed a 2D-E method applicable for protein analysis of Glycyrrhiza uralensis callus. |
