Effects of different interstocks on physiological characteristics and MdPIP1-1 gene expression of grafted apple trees under drought stress

Abstract:
      To explore the effects of different interstocks on drought resistance of Tianhong 2 apple trees, the two\|year\|old Tianhong 2/Huang 6/M.hupehensis,Tianhong 2/SH40/M.hupehensis, Tianhong 2/GM256/M.hupehensis potted apple trees were used as test materials to study effects of different interstocks on drought injury index and related physiological indexes and relative expression of aquaporin gene MdPIP1-1. The results showed that as drought stress time prolonged, soil water content and Fv/Fm decreased, and the drought injury index and O-2 production rate of the three interstock trees increased. And the activities of SOD and POD increased first and then decreased.The ranges of each index of the three rootstock combinations were different. On the 9th day of drought, the drought injury index of Tianhong 2/GM256/M.hupehensis reached the highest level of 91.67%, and its Fv/Fm decreased significantly by 11.47% when compared with normal water supply.The drought injury index of Tianhong 2/Huang 6/M.hupehensis was only 29.55%, and the decrease in Fv/Fm was the smallest. During the drought stress period, the expression of MdPIP1-1 gene in three interstock trees increased first and then decreased. The peak expression of Huang 6 appeared on the 7th day, and the expression of GM256 and SH40 appeared on the 5th day.In general, the drought resistance of the three rootstock combinations was in the order of Tianhong 2/Huang 6/M.hupehensis>Tianhong 2/SH40/M.hupehensis>Tianhong 2/GM256/M.hupehensis.
