In this study, ‘Longyou 7’ and ‘Tianyou 4’ were treated with 400 μmol·L-1 colchicine (Col) and 50 μmol·L-1 taxol (Tax) for 12 h and 24 h at 20℃ and 0℃ and followed by the sample liquid chromatography and quantitative PCR to determine the effects of microtubule reagent on endogenous hormones and cold\|tolerance related genes in seedling winter rapeseed. The results showed that the contents of ‘Longyou 7’ and ‘Tianyou 4’, ABA, IAA, GA3 and ZT decreased significantly after adding Col, and the levels of ABA, IAA, GA3 and ZT increased significantly after Tax. After adding Col, compared to the 0℃ treatment for 12 h, when the 0℃ was processed for 24 h, the expressions of ‘Longyou 7’ α-tubulin, β-tubulin, MKK2, ICE1, CBF1, and BrAFP1 were upregulated, ‘Tianyou 4’ α-tubulin, β-tubulin and CBF1 were upregulated, however, the expression level of MKK2, ICE1 and BrAFP1 was downregulated; compared to the 20℃treatment for 12 h, at 20℃ treatment for 24 h, the expressions of ‘Longyou 7’ and ‘Tianyou 4’ α-tubulin, β-tubulin, MKK2, ICE1, CBF1, and BrAFP1 were upregulated. After adding Tax, compared to the 0℃ treatment for 12 h, when the 0℃ was processed for 24 h, the expressions of ‘Longyou 7’ α-tubulin, β-tubulin, ICE1, CBF1 and BrAFP1 were downregulated. The expression of ‘Tianyou 4’ α-tubulin, β-tubulin, ICE1, CBF1 and BrAFP1 was upregulated; compared to the 20℃ treatment for 12 h, at 20℃ treatment for 24 h, the expressions of ‘Longyou 7’ and ‘Tianyou 4’ α-tubulin, β-tubulin, MKK2, ICE1, CBF1, and BrAFP1 were downregulated. This showed that microtubule regulating reagent affected the endogenous hormone content and the expression of cold resistance genes in cabbage winter rape. |
