Effects of exogenous hydrogen sulfide on photosynthetic and physiological characteristics of kidney bean seedlings under drought stress

Abstract:
      To study the effects of exogenous hydrogen sulfide (H2S) on photosynthetic and physiological characteristics of kidney bean seedlings under drought stress, a pot experiment was adopted using the kidney bean 2242. Water controlled by weighing and normal water supply was used as control. Sodium hydrosulfide (NaHS) was used as H2S donor to spray at 5 concentrations. The results showed that photosynthesis (Pn, Tr, Gs and Ci) were decreased under drought stress, and the decrease was between 48.79% and 63.72%; antioxidant enzyme (SOD, POD, CAT and APX) activities were activated and the increase was between 22.00% and 78.28%; O-2, H2O2, MDA and REC were increased, the increase was between 91.11% and 185.47%; and osmotic regulation substances (Pro and SS) were increased, the increase was between 30.98% and 55.55%. when the days of rewater increased, various indicator values all increased. After applying exogenous NaHS, comparing with MD (Drought stress+0 mmol·L-1 NaHS), photosynthetic efficiency, antioxidant enzyme activities and osmotic regulation substances were increased first and then decreased and the maximum increases were 39.18%~89.69%, 28.40%~94.63% and 15.03%~86.12% at 0.2 mmol·L-1 respectively. Membrane peroxidation were decreased first and then increased, and the maximum decrease was 19.91%~48.37% at 0.2 mmol·L-1 concentration. Correlation analysis showed that the same kind indicators were significantly positive correlation,and the correlation coefficient was shown as antioxidant enzyme activities>photosynthetic efficiency, membrane peroxidation>osmotic regulation substances. There was the strongest positive correlation between antioxidant enzyme activities and osmotic regulation substances among different kinds of indicators. In conclusion, exogenous H2S can alleviate the adverse effects under drought stress on the photosynthetic and physiological characteristics in kidney bean seedlings, reduce the damage to plants and enhance the ability to resist drought. When the concentration of NaHS solution was 0.2 mmol·L-1, it had the most significant effect on alleviating drought stress of kidney bean seedling.
