Genome\|wide identification of CHS gene family in Brassica napus L. and functional analysis of BnCHS04 in response to cold stress

Abstract:
      The CHS gene family in Brassica napus L. was systematically analyzed and investigated. Through bioinformatics analysis, nine BnCHS genes were identified across the B. napus genome and distributed across seven chromosomes. Among these, seven BnCHS genes comprised two exons and one intron. To further elucidate the functional roles of these genes in cold stress responses, their expression levels were analyzed during seedling\|stage cold stress in the extreme cold\|tolerant rapeseed cultivar ‘GZ Hui’ and the extreme cold\|sensitive cultivar ‘10B’. The results showed that five genes—BnCHS02, BnCHS03, BnCHS04, BnCHS07, and BnCHS08—participate in cold stress response. Notably, BnCHS04 is located in the main QTL of cold resistance qFDI.C02.1, indicating its role as a key gene involved in cold tolerance regulation. To further validate the function of BnCHS04, the gene was cloned from the extreme cold\|tolerant rapeseed cultivar ‘GZ Hui’ and heterologously overexpressed in Arabidopsis. Under cold acclimation low\|temperature stress, the percentage of dead leaves of the two overexpressing lines were 12.59% and 10.37%, respectively, while the wild type exhibited a rate of 26.30%. The relative electrical conductivity (REC) for the two overexpressing lines were 0.34 and 0.26, respectively, compared to 0.49 for the wild type. The percentage of dead leaves and REC values of the overexpressing lines were significantly lower than those of the wild type, indicating that BnCHS04 significantly enhanced cold tolerance in Arabidopsis after cold acclimation, confirming its role in strengthening plant cold resistance.
